1,2,3,4-Tetrahydroquinoline: A Versatile Nucleus in the New Millennium

Tetrahydroquinoline nucleus containing molecules are well known for their presence in natural products mainly in alkaloids, which constitute the active class of compounds possessing wide spectrum of biological activities. Tetrahydroquinoline skeleton is often used for the design of several synthetic compounds with diverse pharmacological properties. An immense wide range of pharmacological utility of such nucleus has inspired many synthetic and medicinal chemists to develop novel strategies for their synthesis, which remains an important and challenging task. In the present review, we lay emphasis on the recent advances along with classical methodologies for the direct synthesis of 1,2,3,4-tetrahydroquinoline scaffold mainly using non-heterocyclic moiety as starting material. Apart from the various synthetic methodologies, we also aim to summarize the spectrum of important biological activities like antimicrobial, antiviral, antifungal, antimalarial, antileishmanial, antitubercular, antidiabetic, antithrombotic and anticancer exhibited by 1,2,3,4-tetrahydroquinoline analogues.
